Overview

Hexarelin is a synthetic hexapeptide (His-D-2-methyl-Trp-Ala-Trp-D-Phe-Lys-NH₂) and a potent agonist of the ghrelin / growth-hormone-secretagogue receptor (GHS-R1a), structurally related to GHRP-6. It is supplied as a stable lyophilized peptide.

In research it is studied both as a strong stimulus of growth-hormone (GH) release and, separately, for cardiovascular effects that appear independent of GH — an unusual dual interest for a secretagogue. It is offered strictly as a research material, and the studies cited below describe preclinical and clinical-pharmacology findings only.

Ghrelin / GHS-R1a receptor agonism and GH release

Preclinical / in-vitro

Hexarelin binds GHS-R1a to trigger potent GH release from the pituitary. In model systems its GH response is reproducible and can be modulated by sex-steroid status, making it a well-characterised probe of the GH axis.

CD36 binding and cardiovascular signalling

Preclinical / in-vitro

Beyond the ghrelin receptor, hexarelin also binds the scavenger receptor CD36, identified as a cardiac receptor through which it is studied for cardioprotective effects independent of GH release. This second receptor is a distinctive focus of hexarelin research.

In prepubertal short-statured children, hexarelin produced reproducible GH secretion, and priming with testosterone or ethinyl estradiol significantly enhanced the GH response while oxandrolone did not.
