Overview

Ipamorelin is a synthetic pentapeptide (Aib-His-D-2-Nal-D-Phe-Lys-NH₂) that acts as an agonist of the ghrelin / growth-hormone-secretagogue receptor (GHS-R1a). It was developed as a highly selective secretagogue and is supplied as a stable lyophilized peptide.

In research it is studied as a tool for stimulating growth-hormone (GH) release through the ghrelin-receptor pathway, distinct from growth-hormone-releasing hormone (GHRH). Its defining research interest is selectivity: unlike earlier peptides in this class, it releases GH in model systems with little effect on cortisol, ACTH, or prolactin, making it useful for isolating GHS-R1a signalling. It is offered strictly as a research material, and the studies cited below describe preclinical findings only.

Ghrelin / GHS-R1a receptor agonism

Preclinical / in-vitro

The growth-hormone-secretagogue receptor GHS-R1a, the target of the hormone ghrelin, drives pulsatile GH release from the pituitary. Ipamorelin binds this receptor as an agonist, and in pituitary-cell and animal models it stimulates GH secretion, which is why it is used to probe ghrelin-receptor signalling independently of GHRH.

Selectivity for GH release

Preclinical / in-vitro

A central research feature of ipamorelin is its selectivity: in model systems it releases GH without the increases in ACTH, cortisol or prolactin seen with less selective secretagogues, even at high relative doses. This clean profile is the main reason it is favoured as a research probe of the GH axis.

Growth-hormone axis and downstream endpoints

Preclinical / in-vitro

Sustained GH signalling feeds into IGF-1 production and downstream tissue endpoints such as bone. Ipamorelin has therefore been studied in longer-term animal models examining body weight and bone mineral content as readouts of GH-axis activation.

In rat pituitary cells and in rats and swine, ipamorelin selectively released growth hormone without affecting ACTH or cortisol even at 200-fold the GH ED50, which the authors described as GHRH-like selectivity for a GHS-receptor agonist.
