Overview
Melanotan-1 (afamelanotide) is a synthetic linear analog of α-melanocyte-stimulating hormone (α-MSH), modified for stability. Unlike the broader-acting Melanotan-2, it is relatively selective for the MC1 receptor. It is supplied as a stable lyophilized peptide.
In research it is studied for pigmentation and photoprotection: MC1R activation on melanocytes drives production of the dark pigment eumelanin, which absorbs and scatters ultraviolet light. It is offered strictly as a research material, and the study cited below describes clinical-pharmacology findings only.
Mechanisms of interest in research
MC1 receptor agonism and eumelanin synthesis
Melanotan-1 activates the MC1 receptor on melanocytes, switching pigment production toward eumelanin. This melanogenic action is the central mechanism studied and underlies its interest in photoprotection research.
Photoprotection models
Because eumelanin absorbs ultraviolet radiation, increased eumelanin is examined for reduced phototoxicity, including in light-sensitivity conditions.
